- Dentin sialophosphoprotein (DSPP) is a precursor protein that is cleaved into two functional proteins: dentin sialoprotein (DSP) and dentin phosphoprotein (DPP).
- DSP is implicated in dentinogenesis, the process of dentin formation, although its exact mechanism is not fully characterized.
- DPP is highly acidic and binds calcium ions, facilitating the initial mineralization of dentin matrix collagen and regulating the size and shape of hydroxyapatite crystals.
- DSPP is secreted into the extracellular matrix of developing teeth, where it contributes to biomineralization.
- Tissue expression is restricted to teeth: DPP is synthesized by odontoblasts and transiently expressed by pre-ameloblasts.
- Post-translational modifications include glycosylation, phosphorylation, and sialic acid addition (making DSP a sialoprotein).
- Mutations in DSPP are associated with non-syndromic deafness (DFNA39) and dentinogenesis imperfecta.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 16–230 of human DSPP, corresponding to the DSP region, so the antibody is expected to recognize both full-length DSPP and the cleaved DSP product, but not DPP.
- For Western blot (WB), the predicted molecular weight of full-length DSPP is ~131 kDa; however, cleavage products (e.g., DSP ~95 kDa) may also be detected. Validate in relevant sample lysates such as human dental pulp or odontoblast cell lines.
- For immunofluorescence on paraffin-embedded sections (IF-P), consider antigen retrieval and permeabilization steps, as the target is secreted and may be cross-linked in the extracellular matrix.
- In ELISA, the antibody can be used as a capture or detection reagent; titrate to optimize signal-to-noise ratio in the intended sample matrix.
