Biological Background: Desmoglein-3 Function and Localization

  • Desmoglein-3 (DSG3), also known as 130 kDa pemphigus vulgaris antigen (PVA) and cadherin family member 6 (CDHF6), is a calcium-binding transmembrane glycoprotein of the cadherin superfamily, primarily involved in desmosome-mediated cell-cell adhesion.

  • DSG3 is a core component of desmosome cell-cell junctions and is required for positive regulation of cellular adhesion and for adherens and desmosome junction assembly in response to mechanical force in keratinocytes. Related references: PMID:31835537

  • Essential for desmosome-mediated cell-cell adhesion of cells surrounding the telogen hair club and the basal layer of the outer root sheath epithelium, thereby anchoring telogen hairs in the hair follicle. Related references: PMID:9701552

  • Required for the maintenance of the epithelial barrier by promoting desmosome-mediated intercellular attachment of suprabasal epithelium to basal cells (by similarity).

  • Plays a role in stabilizing the protein levels of desmosome plaque components DSP, JUP, PKP1, PKP2, and PKP3. Related references: PMID:22294297

  • Subcellularly, DSG3 localizes to the cell membrane, desmosomes, tight junctions, and cytoplasm. It is expressed in the basal and spinous layers of epidermis, skin, mucosa, esophageal mucosa, hair follicle outer root sheath, tongue, and tonsil.

  • The immunogen spans amino acids 50–615 of human DSG3, covering the extracellular cadherin repeat domains; this design may contribute to specific recognition of native protein conformations.
  • For IHC-P, standard antigen retrieval protocols (e.g., heat-induced epitope retrieval in citrate buffer, pH 6.0) may be applied; always include appropriate positive and negative tissue controls.
  • For ELISA, the recombinant immunogen protein can serve as a tool for assay optimization and as a positive control; perform endpoint titration to determine optimal antibody dilution.
  • This antibody reacts with human DSG3; cross-reactivity with other desmoglein or cadherin family members has not been fully characterized — independent validation is recommended for multiplex panels.
