Biological Background: DSG1 Function and Localization
- Component of desmosome cell-cell junctions, positively regulating cellular adhesion (PubMed:19717567). Related references: PMID:19717567
- Promotes differentiation of keratinocytes by interacting with ERBIN, reducing RAS/ERK signaling through SHOC2 competition (PubMed:23524970). Related references: PMID:23524970
- Mediates interaction between desmosome and COP9 signalosome, promoting keratinocyte differentiation via deneddylation of EGFR and reducing EGFR stabilization (PubMed:28891468). Related references: PMID:28891468
- Critical for epidermal integrity and skin barrier function (By similarity).
- Required for esophageal epithelial barrier integrity and correct intercellular spacing (PubMed:24220297). Related references: PMID:24220297
- Required for recovery of epidermal architecture and correct differentiation marker expression following UVB exposure (PubMed:24594668). Related references: PMID:24594668
- Subcellularly located at cell membrane, desmosome, cytoplasm, and nucleus.
- Tissue specificity: expressed in all suprabasal epidermal layers, highest in granular layer, and in suprabasal esophageal epithelium (PubMed:24220297). Related references: PMID:24220297
Experimental Guidance and Technical Tips
- This polyclonal antibody, raised against a fragment corresponding to amino acids 800–1049 of human DSG1, may be suitable for Western blot (predicted band ~114 kDa), immunofluorescence on paraffin-embedded sections (IF-P), and ELISA.
- For Western blot, consider using reducing conditions and validate detection in your sample system, given reported cross-reactivity with both human and rat DSG1.
- For IF-P, antigen retrieval may be required; optimize based on your tissue and fixation protocol.
- As with any polyclonal antibody, lot-to-lot consistency should be verified for critical applications.
