Biological Background: Desmocollin-2 (DSC2) Function and Localization
- Desmocollin-2 (DSC2), also known as cadherin family member 2 (CDHF2), desmocollin-3 (DSC3), and desmosomal glycoprotein II/III (DGII/III), is a calcium-dependent cell adhesion protein and a core component of desmosomes.
- It positively regulates cellular adhesion by promoting incorporation of desmoglein-2 (DSG2) into intercellular junctions and mediating interaction with the intermediate filament cytoskeleton via modulation of desmoplakin (DSP) phosphorylation. Related references: PMID:33596089
- DSC2 is critical for maintaining intestinal epithelial barrier function and intercellular adhesion strength. Related references: PMID:33596089
- It promotes wound healing in the intestinal mucosa by enhancing epithelial cell migration and participates in mechanotransduction between cells and the extracellular matrix. Related references: PMID:31967937
- Subcellular localization is predominantly at the cell membrane within desmosome cell junctions.
- Tissue-specific expression includes intercalated disks in the heart (colocalized with CDH2) and intestinal mucosal cells, as confirmed at the protein level. Related references: PMID:23863954 PMID:33784018
- The protein features cadherin repeat domains, calcium-binding sites, a transmembrane region, and is subject to post-translational modifications including cleavage, phosphorylation, and glycosylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 520–690 within the extracellular domain of human DSC2 (NP_077740.1). This region may offer specificity for DSC2, but cross-reactivity with closely related cadherins should be assessed experimentally.
- Validated applications include Western blot (WB) and ELISA. For WB, the expected molecular weight is 100 kDa. Consider using human heart or intestinal tissue lysates as positive controls, and optimize primary antibody dilution and blocking conditions in your specific assay context.
- As a rabbit polyclonal antibody, it may detect multiple isoforms or modified forms; careful validation of band patterns is recommended.
