Biological Background: DROSHA Function and Localization

  • DROSHA (Ribonuclease 3) is a nuclear protein also known as RNASEN, RN3, or RNase III, encoded by the DROSHA gene. It is a member of the ribonuclease III family of double-stranded RNA-specific endoribonucleases.
  • As the catalytic subunit of the microprocessor complex, DROSHA processes primary miRNA transcripts (pri-miRNAs) into ~70-nucleotide precursor miRNAs (pre-miRNAs) in the nucleus, initiating the canonical miRNA biogenesis pathway.
  • It cleaves both the 3' and 5' strands of the stem-loop structure in pri-miRNAs at a defined distance from the junction, releasing hairpin pre-miRNAs for further processing by DICER in the cytoplasm.
  • Beyond miRNA maturation, DROSHA participates in pre-ribosomal RNA processing and plays a role in the formation of GW/P-bodies, linking it to RNA metabolism and gene silencing.
  • Subcellularly, DROSHA predominantly localizes to the nucleus, with enrichment in the nucleolus, but can also be detected in the cytoplasm under certain conditions.
  • The protein is ubiquitously expressed and subject to alternative splicing and post-translational modifications including phosphorylation, which may regulate its activity.
  • Keywords associated with DROSHA include: Endonuclease, Hydrolase, RNA-binding, Magnesium/Manganese metal-binding, and RNA-mediated gene silencing.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from the N-terminal region (aa 1-100). For applications like Western blot, the full-length protein migrates at ~159 kDa; ensure adequate transfer and use appropriate molecular weight markers.
  • For immunofluorescence/ICC, fixation and permeabilization conditions should be optimized as the target is predominantly nuclear; consider using a nuclear counterstain to confirm localization.
  • ELISA performance can be verified using recombinant DROSHA protein or synthetic peptide; titration of the antibody is recommended.
  • Verify reactivity in your specific human-derived sample system, as expression levels may vary.
