Biological Background: DRAP1 Function and Localization

  • DRAP1 (Dr1-associated protein 1), also known as negative cofactor 2-alpha (NC2-alpha), is a transcriptional corepressor that forms a heterodimer with DR1.
  • The DR1/DRAP1 heterodimer binds to TATA-box binding protein (TBP) to repress both activated and basal transcription of class II genes by inhibiting the assembly of transcription-competent complexes (preventing association of TFIIA and/or TFIIB with TBP).
  • DRAP1 can also bind DNA independently, suggesting an additional role in gene regulation.
  • The protein localizes to the nucleus, consistent with its function in transcriptional regulation.
  • DRAP1 undergoes alternative splicing and phosphorylation, as indicated by UniProt keywords.
  • It is ubiquitously expressed, with highest levels in adult testis, heart, skeletal muscle, pancreas, and brain, as well as fetal brain, liver, and kidney.
  • The predicted molecular weight of DRAP1 is approximately 22 kDa (UniProt: 22350 Da).

Experimental Guidance and Technical Tips

  • The immunogen (aa 50–150) encompasses the conserved histone fold domain, which may contribute to the observed cross-reactivity with mouse and rat DRAP1.
  • For Western blot, expect a band around 22 kDa; nuclear extracts or whole-cell lysates from human, mouse, or rat cells/tissues are suitable starting samples.
  • In ELISA, consider using the antibody for antigen capture or detection, and validate with appropriate positive (e.g., recombinant DRAP1) and negative controls.
  • As DRAP1 is a nuclear protein, ensure proper sample preparation (e.g., nuclear extraction or lysis buffer optimization) to maximize detection sensitivity.
  • As a polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative applications.
