Biological Background: DR4 Function and Localization
- Receptor identity: TNFRSF10A (Death receptor 4, TRAIL-R1) is a member of the tumor necrosis factor receptor superfamily, characterized by an extracellular domain containing cysteine-rich repeats.
- Apoptosis induction: DR4 serves as a receptor for the cytotoxic ligand TNFSF10/TRAIL. Upon ligand binding, the adapter molecule FADD recruits caspase-8 to form the death-inducing signaling complex (DISC), leading to proteolytic activation of caspase-8 and initiation of the apoptotic cascade (PubMed:19090789, PubMed:26457518, PubMed:38532423). - NF-kappa-B activation: In addition to apoptosis, DR4 promotes the activation of the NF-kappa-B signaling pathway, contributing to inflammatory and survival responses (PubMed:9430227). Related references: PMID:9430227
- Subcellular localization: The receptor is primarily localized to the cell membrane and membrane rafts, but it can also be detected in the cytoplasm/cytosol.
- Tissue expression: DR4 is widely expressed, with notably high levels in the spleen, peripheral blood leukocytes, small intestine, thymus, and certain cancer cell lines (K-562, MCF-7), as well as activated T-cells.

Experimental Guidance and Technical Tips
- The immunogen covers amino acids 124–232 within the extracellular domain of human DR4; thus, the antibody likely recognizes epitopes in the ligand-binding region and may preferentially detect the native conformation.
- For flow cytometry (FC), staining of live, non-permeabilized cells is expected to label cell-surface DR4, while intracellular detection may require permeabilization.
- In ELISA applications, the antibody can be used for detection of recombinant or native DR4; sandwich immunoassays may benefit from pairing with a second antibody targeting a non-overlapping epitope.
- As a rabbit monoclonal antibody, it provides batch-to-batch consistency; optimal working concentrations should be determined empirically for each assay system.
- Cross-reactivity is reported for human DR4 only; performance with DR4 from other species has not been characterized.
