Biological Background: DPYD Function and Localization

  • DPYD (dihydropyrimidine dehydrogenase) is the initial and rate-limiting enzyme of the pyrimidine catabolic pathway, catalyzing the NADPH-dependent reduction of uracil and thymine to dihydrouracil and dihydrothymine, respectively.
    Related references: PMID:1512248
  • It is also responsible for the metabolic inactivation of the chemotherapeutic prodrug 5-fluorouracil (5-FU), converting it to dihydrofluorouracil and thereby modulating drug efficacy and toxicity.
    Related references: PMID:1512248
  • Alternative designations include dihydrothymine dehydrogenase, dihydrouracil dehydrogenase, DPD, and DHPDHASE.
  • The enzyme is widely expressed, with the highest catalytic activity found in liver and peripheral blood mononuclear cells.
  • DPYD localizes predominantly to the cytoplasm.
  • The protein contains multiple cofactors essential for its oxidoreductase function, including 4Fe-4S iron-sulfur clusters, FAD, FMN, and NADP.

Experimental Guidance and Technical Tips

  • The immunogen encompasses the N-terminal region (residues 1–173) of human DPYD; therefore, the antibody may detect multiple splice variants. Validate reactivity in your target tissue or cell line.
  • For Western blotting, a specific band near 111 kDa is anticipated. Use appropriate gel percentage (e.g., 8–10% SDS-PAGE) and optimize blocking/dilution conditions to minimize non-specific binding.
  • Include positive controls such as human liver lysates or mouse tissues rich in DPYD activity to confirm assay performance.
  • For ELISA applications, test direct binding or explore sandwich pair compatibility with a matched detection antibody if quantitative purposes are required.
