Biological Background: DPH2 Function and Localization

  • DPH2 encodes subunit 2 of 2-(3-amino-3-carboxypropyl)histidine synthase, also known as diphthamide biosynthesis protein 2, essential for the unique post-translational modification of elongation factor 2.
  • Required for the first step of diphthamide biosynthesis, a post-translational modification of histidine which occurs in elongation factor 2. Related references: PMID:32576952
  • Together with DPH1, transfers a 3-amino-3-carboxypropyl group from S-adenosyl-L-methionine to the target histidine; the reaction depends on the DPH3 and NADH-dependent reductase system.
  • Facilitates reduction of the catalytic iron-sulfur cluster found in the DPH1 subunit, underlining its role in enzyme activation.
  • Strongly expressed in skeletal muscle; moderately in heart, small intestine, liver, pancreas, testis, and colon; weakly in brain, placenta, kidney, spleen, thymus, prostate, ovary, and lymphocytes.
  • Post-translational modifications include acetylation and phosphorylation; iron-sulfur cluster binding is critical for function.
  • Mutations in DPH2 are associated with developmental disorders featuring dwarfism, ectodermal dysplasia, hypotrichosis, and intellectual disability.
