Biological Background: DP1/TFDP1 Function and Localization
- DP1, also known as Transcription factor Dp-1, is the protein product of the TFDP1 gene and serves as a critical dimerization partner for E2F transcription factors.
- It stimulates E2F-dependent transcription by binding cooperatively with E2F family members to the E2 recognition site (5'-TTTC[CG]CGC-3') found in promoters of genes involved in cell cycle regulation and DNA replication. Related references: PMID:7739537, PMID:8405995
- The E2F1:DP complex mediates both cell proliferation and apoptosis, and DP1 blocks adipocyte differentiation by repressing CEBPA binding to target gene promoters. Related references: PMID:20176812
- Subcellular localization is primarily nuclear, but the protein is also detected in the cytoplasm, suggesting dynamic shuttling between these compartments.
- Expression is highest in muscle, with moderate levels in brain, placenta, liver, and kidney, low levels in lung and pancreas, and no detection in heart.

- The immunogen is a synthetic peptide spanning amino acids 311–410 near the C-terminus; the antibody is expected to detect full-length DP1, but isoforms missing this epitope may not be recognized. Validate in your sample system.
- For Western blotting, a band around 45 kDa is anticipated. Include appropriate positive controls (e.g., HeLa or HEK293 lysates) and loading controls.
- In IHC-P, optimize antigen retrieval and consider that nuclear staining is predominant, though cytoplasmic signal may also appear due to shuttling.
- This antibody shows cross-reactivity with human and rat; for other species, verify target sequence homology before use.
