Biological Background: Doublecortin Function and Localization
- Doublecortin (DCX) is a microtubule-associated protein essential for neuronal dispersion and cortex lamination during cerebral cortex development. It likely acts by competing with DCLK1 kinase in binding to a target protein, potentially participating in a calcium-dependent signal transduction pathway crucial for neuronal migration.
- DCX is part of overlapping but distinct signaling pathways with PAFAH1B1/LIS-1, both promoting neuronal migration.
- Also known as Doublin and Lissencephalin-X, mutations in DCX are associated with lissencephaly and epilepsy.
- The protein localizes to the cytoplasm and neuron projections.
- It is highly expressed in neuronal cells of the fetal brain, particularly in the cortical plate, intermediate zone, and ventricular zone. In adults, expression is prominent in the frontal lobe of the brain but very low elsewhere, and absent from non-neuronal tissues such as heart, placenta, lung, liver, skeletal muscle, kidney, and pancreas.
- DCX undergoes phosphorylation and ubiquitination (Ubl conjugation), and alternative splicing generates multiple isoforms. It contains repeat domains and is classified as a developmental protein involved in neurogenesis and differentiation.
