Biological Background: DOG-1/TMEM16A Function and Localization
- ANO1 encodes anoctamin-1, a calcium-activated chloride channel (CaCC) that plays central roles in transepithelial anion transport and smooth muscle contraction. Related references: PMID:20056604, PMID:22178883, PMID:22946059
- The channel is indispensable for the pacemaker activity of interstitial cells of Cajal (ICCs) in the gastrointestinal tract, thereby governing gut motility.
- In airway epithelial cells, ANO1 contributes substantially to basal and stimulated chloride conductance and is critical for tracheal cartilage development.

- The immunogen (aa 40–210) corresponds to the N-terminal region; detection of certain splice isoforms may vary, so verify isoform-specific recognition if needed.
- When performing ELISA, titrate the antibody to determine optimal working concentrations and confirm specificity against recombinant ANO1 protein or overexpression cell lysates.
