Biological Background: DNMT3L Function and Localization
- DNMT3L is a catalytically inactive regulatory factor of DNA methyltransferases that can either promote or inhibit DNA methylation depending on the context.
- Essential for the function of DNMT3A and DNMT3B: activates DNMT3A and DNMT3B by binding to their catalytic domain (PubMed:17687327). Related references: PMID:17687327
- It accelerates the binding of DNA and S-adenosyl-L-methionine (AdoMet) to the methyltransferases and dissociates from the complex after DNA binding (PubMed:17687327). Related references: PMID:17687327
- DNMT3L recognizes unmethylated histone H3 lysine 4 (H3K4me0) and induces de novo DNA methylation by recruitment or activation of DNMT3 (PubMed:17687327). Related references: PMID:17687327
- In germ cells, DNMT3L is required for the methylation of imprinted loci together with DNMT3A, and in male germ cells, it specifically methylates retrotransposons to prevent their mobilization.
- In embryonic stem cells, DNMT3L both promotes DNA methylation of housekeeping genes and inhibits methylation at bivalent gene promoters by interacting with the EZH2 component of the PRC2/EED-EZH2 complex, preventing DNMT3A/B recruitment.
- DNMT3L promotes differentiation of ESCs into primordial germ cells by inhibiting DNA methylation at the RHOX5 promoter, thereby activating its expression.
- Subcellularly, DNMT3L localizes predominantly to the nucleus, consistent with its role in chromatin modification.
- It is expressed at low levels in several tissues including testis, ovary, and thymus, and belongs to the zinc-finger protein family, featuring metal-binding motifs.
Experimental Guidance and Technical Tips
- Consider performing nuclear fractionation to enrich DNMT3L for Western blot detection, as it localizes to the nucleus.
- For IHC-P, antigen retrieval may be necessary; validate in tissues with known low expression such as testis or ovary.
- The immunogen sequence is conserved among human, mouse, and rat; cross-reactivity is predicted but should be confirmed experimentally.
- The antibody detects a band around 44 kDa; a pre-stained protein ladder is recommended for accurate size estimation.
- Peptide blocking with the immunogen can serve as a specificity control for both WB and IHC-P assays.
