Biological Background: DNMT3B Function and Localization
- Encodes DNA (cytosine-5)-methyltransferase 3B (DNMT3B), also known as DNA methyltransferase HsaIIIB (M.HsaIIIB), with multiple isoforms; isoforms 4 and 5 lack conserved methyltransferase motifs and are likely non-functional.
- Essential for genome-wide de novo DNA methylation during development, establishing methylation patterns coordinated with histone methylation (By similarity).
- Preferentially methylates nucleosomal DNA within the nucleosome core region and may function as a transcriptional co-repressor by associating with CBX4 or ZHX1 independently of its catalytic activity (By similarity).
- Required for DUX4 silencing in somatic cells, highlighting its role in gene repression (PubMed:27153398). Related references: PMID:27153398
- Subcellularly localized to the nucleus, consistent with its function in chromatin modification.
- Ubiquitously expressed with highest levels in fetal liver, heart, kidney, and placenta; specific isoforms show tissue-restricted patterns (e.g., isoform 5 is testis-specific).
- Post-translationally modified by phosphorylation, citrullination, and ubiquitination; contains zinc-finger domains and is a S-adenosyl-L-methionine-dependent methyltransferase.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 345-475 of human DNMT3B, a region within the catalytic methyltransferase domain; this suggests the antibody may recognize both full-length and truncated isoforms that retain this region.
- For Western blot (WB), the predicted molecular weight of the canonical isoform is 96 kDa, but other isoforms and post-translational modifications may produce additional bands; consider using nuclear extracts from human, mouse, or rat tissues/cell lines.
- For immunofluorescence (IF/ICC), nuclear staining is expected; validate with appropriate nuclear counterstains and controls in relevant cell systems.
- For ELISA, perform standard titration and specificity verification using the recombinant immunogen or full-length protein as positive control.
- Due to the polyclonal nature, batch-to-batch consistency should be verified with appropriate positive and negative controls in the intended application.
