Biological Background: DNMT3B Function and Localization
- DNMT3B is a DNA (cytosine-5)-methyltransferase that catalyzes de novo DNA methylation, establishing methylation patterns critical for embryonic development and cellular differentiation.
- It preferentially methylates nucleosomal DNA within the core region and may coordinate DNA methylation with histone modifications to regulate chromatin structure and gene expression.
- Multiple alternatively spliced isoforms exist; isoforms 1, 3, and possibly 2 are functional, while isoforms 4 and 5 lack conserved methyltransferase motifs and are likely catalytically inactive.
- Beyond its catalytic role, DNMT3B functions as a transcriptional corepressor by interacting with CBX4 and ZHX1, mediating gene silencing independently of methylation activity.
- In association with DNMT1 and the factor CTCFL/BORIS, DNMT3B modulates histone H3K4 and H3K9 dimethylation at the BAG1 promoter, contributing to transcriptional activation.
- DNMT3B is required for silencing of the DUX4 gene in somatic cells; its dysregulation is linked to facioscapulohumeral muscular dystrophy type 4 (FSHD4). Related references: PMID:27153398
- Localized to the nucleus, DNMT3B is widely expressed, with high levels in fetal liver, heart, kidney, and placenta, and lower levels in adult tissues such as brain and skeletal muscle. Isoform 1 is absent in brain, skeletal muscle, and PBMC.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 206–360 of human DNMT3B, a region containing part of the PWWP domain; verify expression of functional isoforms in your sample system to ensure detection.
- For Western blot (WB), expect a band near 96 kDa; validate specificity with DNMT3B knockout or knockdown controls, as some isoforms may migrate at different sizes.
- For ChIP, carefully optimize crosslinking, shearing, and antibody concentration, as DNMT3B is a nuclear chromatin-associated protein; include appropriate negative regions.
- In ELISA, generate a standard curve with the recombinant immunogen to assess linearity; the antibody may also be used for capture or detection in sandwich ELISA with matched pairs.
- Because DNMT3B is expressed at low levels in many tissues, consider nuclear enrichment or high-protein loading for sensitive detection.
- Cross-reactivity with non-human species is not reported; validate in the relevant system if used beyond human samples.
