Biological Background: DNMT3B Function and Localization

  • DNMT3B (DNA (cytosine-5)-methyltransferase 3B) is a key enzyme required for genome-wide de novo DNA methylation, essential for establishing methylation patterns during early development.
  • It preferentially methylates nucleosomal DNA within the core region and can function as a transcriptional co-repressor by interacting with CBX4 and ZHX1, independently of its methyltransferase activity.
  • In a complex with DNMT1 and the transcription factor CTCFL/BORIS, DNMT3B activates BAG1 gene expression through modulation of histone H3 dimethylation at K4 and K9.

  • Alternative splicing generates several isoforms; isoforms 4 and 5 lack critical methyltransferase motifs and are likely non-functional.
  • DNMT3B is exclusively nuclear and is ubiquitously expressed, with highest levels in fetal liver, heart, kidney, and placenta. Isoform 1 is absent in brain, skeletal muscle, and PBMC, while isoform 5 is testis-specific.

  • For Western blotting, the predicted molecular weight is ~96 kDa. Use nuclear extracts or whole-cell lysates from tissues with high DNMT3B expression (e.g., fetal liver or placenta) for optimal detection.
  • This antibody is reported to cross-react with mouse and rat; validate performance in the relevant species and application.
  • For immunohistochemistry (IHC-P), consider testing on fetal tissue sections where DNMT3B is abundant. Antigen retrieval conditions may require optimization.
  • ELISA applications should include proper negative and positive controls to ensure specificity.
