Biological Background: DNMT3A Function and Localization

  • DNMT3A is a de novo DNA methyltransferase required for genome-wide methylation patterning during development. It coordinates DNA and histone methylation, and modifies non-CpG sites in a non-processive manner.

  • May preferentially methylate DNA linker between two nucleosomal cores; inhibited by histone H1 (By similarity).
  • Plays a role in paternal and maternal imprinting and is required for methylation of most imprinted loci in germ cells (By similarity).
  • Acts as a transcriptional corepressor for ZBTB18, recruited to trimethylated H3K36 (H3K36me3), and can repress transcription through HDAC recruitment (By similarity).
  • Exhibits weak auto-methylation activity on Cys-710 in the absence of DNA (By similarity).
