Biological Background: DNMT1 Function and Localization
- DNMT1, also known as DNA methyltransferase HsaI, MCMT, or CXXC9, is a ~183 kDa nuclear protein that serves as the primary maintenance DNA methyltransferase in humans.
- It preferentially methylates hemimethylated CpG dinucleotides, and associates with DNA replication foci during S phase to faithfully copy methylation patterns onto the newly synthesized strand, ensuring epigenetic inheritance. Related references: PMID:17200670, PMID:21745816, PMID:26070743
- During G2 and M phases, DNMT1 remains chromatin-associated to sustain methylation independently of replication, thereby preserving genomic methylation marks across the cell cycle. Related references: PMID:21745816
- DNMT1 also mediates transcriptional repression by directly binding HDAC2 and can coordinate DNA methylation with histone modifications, such as H3K9 dimethylation, in concert with DNMT3B and CTCFL. Related references: PMID:10888872, PMID:18413740

Experimental Guidance and Technical Tips
- The antibody was raised against an N-terminal fragment (aa 1-124) of DNMT1; therefore, it is expected to recognize full-length DNMT1 and possibly N-terminal truncation products.
- When performing western blot, use a 6-8% SDS-PAGE gel and ensure efficient transfer due to the large size of DNMT1 (~183 kDa).
- For immunohistochemistry and immunofluorescence, nuclear permeabilization may be required; consider validating staining specificity with DNMT1 knockdown or knockout controls.
- The antibody cross-reacts with mouse and rat DNMT1, but users should verify performance in each species and tissue type given the ubiquitous but variable expression.
- As a polyclonal antibody, it may exhibit lot-to-lot variability; include a loading control and internal reference standards for quantitative assays.
