Biological Background: Dynamin-2 Function and Localization

  • Dynamin-2 is a member of the dynamin family of large GTPases, encoded by the DNM2 gene, and exists in multiple alternatively spliced isoforms.
  • It catalyzes GTP hydrolysis to drive vesicle scission during clathrin-mediated endocytosis, macropinocytosis, and trafficking from the trans-Golgi network. Related references: PMID:15731758, PMID:19623537, PMID:33713620
  • Dynamin-2 self-assembles into helical polymers around vesicle necks, and GTP hydrolysis triggers disassembly, facilitating membrane fission. Related references: PMID:17636067, PMID:34744632, PMID:36445308
  • It also regulates the actin cytoskeleton by bundling actin filaments and remodeling them through interactions with cortactin (CTTN), affecting stress fiber formation and cell motility. Related references: PMID:19605363
  • In autophagy, dynamin-2 participates in ATG9A vesicle formation and recycling endosome scission to release autophagosomes. Related references: PMID:29437695, PMID:32315611
  • Subcellularly, dynamin-2 localizes to clathrin-coated pits, endosomes, the actin cytoskeleton, centrosomes, the postsynaptic density, and the midbody.
  • The protein is widely expressed, notably in skeletal muscle and peripheral nerves, and mutations are linked to Charcot-Marie-Tooth disease (CMT) and centronuclear myopathy. Related references: PMID:7590285
  • Post-translational modifications include acetylation and phosphorylation, and the protein is subject to alternative splicing.

Experimental Guidance and Technical Tips

  • For Western blot (WB), the predicted molecular weight of dynamin-2 is ~98 kDa; validate detection in your sample system with appropriate controls.
  • In immunofluorescence (IF/ICC), because dynamin-2 localizes to punctate cytoplasmic vesicles and the plasma membrane, consider using permeabilization conditions that preserve membrane structures.
  • This antibody is reported to cross-react with mouse and rat, so it can be used for comparative studies; however, confirm cross-reactivity in your specific experimental setup.
  • For ELISA applications, titrate antibody and antigen concentrations to establish optimal signal-to-noise ratios.
