Biological Background: DNM1 Function and Localization
- Dynamin-1 (DNM1) is a large GTPase with a molecular weight of ~97 kDa (UniProt: 97,408 Da) that plays a central role in membrane fission during endocytosis.
- Catalyzes the hydrolysis of GTP to mediate vesicle scission and participates in multiple endocytic pathways, including clathrin-mediated endocytosis and synaptic vesicle endocytosis (PubMed:15703209, PubMed:20428113, PubMed:29668686). Related references: PMID:15703209 PMID:20428113 PMID:29668686
- Self-assembles into rings and helical polymers around vesicle necks, constricting the membrane; GTP hydrolysis then triggers conformational changes that promote membrane fission (PubMed:30069048, PubMed:7877694, PubMed:9922133, PubMed:19084269). Related references: PMID:30069048 PMID:7877694 PMID:9922133 PMID:19084269
- Regulates early stages of clathrin-mediated endocytosis in non-neuronal cells via dephosphorylation downstream of EGFR signaling, and interacts with DNAJC6 during clathrin-coated vesicle (CCV) formation (PubMed:29668686, PubMed:12791276). Related references: PMID:29668686 PMID:12791276
- Mediates rapid endocytosis (RE), a Ca²⁺-dependent, clathrin- and K⁺-independent process in chromaffin cells; also associates with microtubules and produces force (By similarity).
- Subcellularly localizes to the cell membrane, clathrin-coated pits, cytoplasmic vesicles, presynapse, and chromaffin granules.
- Subject to multiple post-translational modifications, including phosphorylation, methylation, and nitration; alternative splicing generates multiple isoforms. Keywords also highlight involvement in disease variants and epilepsy.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering amino acids 400–490 of human DNM1, a central region. Verify sequence homology if the antibody is intended for use in other species.
- For Western blotting, DNM1 migrates at approximately 97 kDa under denaturing conditions; include appropriate molecular weight markers and validate with positive control lysates from human, mouse, or rat brain or neuronal cell lines.
- For ELISA, consider using the recombinant immunogen or full-length DNM1 as coating antigen; perform checkerboard titration to optimize working concentrations of capture and detection reagents.
- Cross-reactivity is reported for human, mouse, and rat; however, individual experimental systems may require optimization of blocking and antibody dilution conditions.
