Biological Background: DNASE1 Function and Localization

DNASE1 (Deoxyribonuclease I) is a secreted serum endonuclease produced by a variety of exocrine and endocrine organs. It preferentially cleaves protein-free DNA and has been implicated in apoptosis (PubMed:11241278, 2251263, 2277032).

It specifically binds to G-actin and blocks actin polymerization (By similarity).

- The immunogen used is a recombinant fusion protein corresponding to amino acids 30–235 of human DNASE1, covering a large portion of the secreted mature protein. This broad epitope coverage increases the likelihood of recognizing native and denatured forms.
- For Western blot (WB), a band near 31 kDa is expected; working dilutions should be optimized by the end-user.
- For immunofluorescence/immunocytochemistry (IF/ICC), consider using permeabilization agents, as DNASE1 is primarily secreted and may require cell membrane permeabilization for detection.
- Cross-reactivity with mouse and rat was observed; thus, this antibody may be suitable for studies using rodent models. However, validation in the specific sample system is recommended.
