Biological Background: DNAL1 Function and Localization

  • DNAL1 encodes dynein axonemal light chain 1, an essential component of the outer arm dynein (ODA) multisubunit ATPase complex in motile cilia.
  • It plays a critical role in generating force for ciliary motility and governing beat frequency (By similarity).
  • DNAL1 is involved in a mechanosensory feedback mechanism that controls ODA activity by tethering the outer arm dynein heavy chain (DNAH5) to the microtubule within the axoneme (By similarity).
  • This protein is important for ciliary function in the airways and for the cilia that produce nodal flow essential for left-right asymmetry determination (PubMed:21496787). Related references: PMID:21496787
  • DNAL1 localizes to the cytoplasm, specifically to the cilium axoneme cytoskeleton.
  • It is expressed in tissues carrying motile cilia, such as respiratory epithelia, ependyma, and testis.
  • Mutations in DNAL1 are associated with ciliopathies, including primary ciliary dyskinesia (CILD16) and Kartagener syndrome.
  • The protein contains leucine-rich repeats and is subject to post-translational modifications including acetylation and phosphorylation.

Experimental Guidance and Technical Tips

  • Raised against a recombinant fragment (aa 41-190) of human DNAL1; verify epitope recognition if precise mapping is required.
  • For Western blot, a band around 22 kDa is expected; consider using reducing conditions and validate with positive controls such as testis or respiratory epithelial lysates.
  • In immunofluorescence/ICC, target the cilium axoneme; co-stain with an axoneme marker to confirm specificity.
  • For ELISA, the antibody may be suitable as capture or detection reagent; perform checkerboard titrations for optimal pairing.
