Biological Background: DNAL1 Function and Localization
- Encodes dynein axonemal light chain 1 (DNAL1), a component of the outer arm dynein (ODA) in motile cilia.
- Part of multisubunit axonemal ATPase complexes that generate force for cilia motility and regulate beat frequency (By similarity).
- Functions in a mechanosensory feedback mechanism tethering ODA heavy chain DNAH5 to microtubules, modulating ODA activity in response to conformational cues (By similarity).
- Critical for ciliary function in airway epithelia and for generating nodal flow essential for left-right body axis determination. Related references: PMID:21496787
- Localizes to the axoneme of motile cilia within the cytoplasm and cytoskeleton.
- Expressed specifically in tissues with motile cilia, including respiratory epithelia, ependyma, and testis.
- Post-translationally modified by acetylation and phosphorylation; associated with primary ciliary dyskinesia and Kartagener syndrome.
- Contains leucine-rich repeats (LRR) that may facilitate protein-protein interactions within the axonemal dynein complex.
Experimental Guidance and Technical Tips
- The immunogen encompasses amino acids 41–190 of human DNAL1, a region that may be conserved across species, potentially contributing to the observed cross-reactivity with mouse and rat.
- For Western blot, a band around 22 kDa is expected; consider using appropriate protein markers and optimizing SDS-PAGE conditions for low molecular weight targets.
- In ELISA applications, the recombinant protein used for immunization could serve as a positive control; assess antibody titer and specificity in your sample matrix.
- For ciliary protein studies, sample preparation may require specialized extraction methods to enrich axonemal fractions; validate antibody performance in the relevant tissue or cell system.
