Biological Background: DNAJC9 Function and Localization
- DNAJC9, also known as DnaJ homolog subfamily C member 9 or DnaJ protein SB73, is a 30 kDa protein belonging to the DnaJ/Hsp40 family, sharing structural features with J-domain co-chaperones.
- It functions as a dual histone chaperone and heat shock co-chaperone, interacting with histone H3-H4 heterodimers and assisting MCM2 in nucleosome assembly. Related references: PMID:33857403
- DNAJC9 also acts as a co-chaperone for the HSP70 family, including HSPA1A, HSPA1B, and HSPA8, recruiting them to histone substrates to maintain chromatin integrity. Related references: PMID:17182002, PMID:33857403
- It forms a co-chaperone complex with TONSL and recruits histone chaperones ASF1A, NASP, and SPT2, facilitating histone H3-H4 transfer and chromatin assembly. Related references: PMID:33857403
- Subcellularly, DNAJC9 localizes to the nucleus, cytoplasm, and cell membrane, consistent with its roles in chromatin dynamics and heat shock responses.
- The protein is subject to phosphorylation and has been identified in multiple proteomics studies, indicating regulation by post-translational modification.
- DNAJC9 is broadly expressed across tissues including heart, placenta, liver, skeletal muscle, kidney, pancreas, thymus, ovary, colon, and peripheral blood.
Experimental Guidance and Technical Tips
- The immunogen spans nearly the full-length human DNAJC9 protein (aa 1–260), which may provide broad epitope recognition and high sensitivity. Optimize antibody dilution for each application.
- For WB, the expected band is ~30 kDa; include appropriate positive control lysates from human cell lines expressing DNAJC9.
- In IF/ICC, consider fixation/permeabilization conditions that preserve nuclear and cytoplasmic epitopes, and validate with DNA or organelle markers.
- Cross-reactivity is reported for human; if working with other species, verify sequence homology and perform validation experiments.
