Biological Background: DNAJC5 Function and Localization

  • DNAJC5, also known as cysteine string protein (CSP) or ceroid-lipofuscinosis neuronal protein 4 (CLN4), is a member of the DnaJ/Hsp40 chaperone family.
  • Acts as a general chaperone in regulated exocytosis and as a co-chaperone for the SNARE protein SNAP-25.
  • Involved in calcium-mediated control of a late stage of exocytosis and may be important for presynaptic function and calcium-dependent neurotransmitter release.
  • Localizes to the cytoplasm, cell membrane, and cytoplasmic vesicles including chromaffin granule membranes and melanosomes.
  • Expressed in a wide range of tissues: pancreas, kidney, skeletal muscle, liver, lung, placenta, brain, and heart.
  • Undergoes post-translational modifications such as palmitoylation, phosphorylation, and acetylation.
  • Mutations in DNAJC5 are associated with neuronal ceroid lipofuscinosis (CLN4 disease), a neurodegenerative disorder.
