Biological Background: DNAJC19 Function and Localization
- DNAJC19, also known as Mitochondrial import inner membrane translocase subunit TIM14 or DnaJ homolog subfamily C member 19, is a 12 kDa protein encoded by the DNAJC19 (TIM14, TIMM14) gene in humans.
- It functions as a mitochondrial co-chaperone that forms a complex with prohibitins to regulate cardiolipin remodeling.
- DNAJC19 is a component of the PAM complex, which is required for the ATP-dependent translocation of transit peptide-containing proteins from the inner membrane into the mitochondrial matrix.
- The protein localizes to the mitochondrion inner membrane and is ubiquitously expressed in human tissues.
- Keywords associated with DNAJC19 include Acetylation, Alternative splicing, Cardiomyopathy, Chaperone, Membrane, Mitochondrion, Mitochondrion inner membrane, Phosphoprotein, Protein transport, Translocation, Transmembrane, and Transport.
Experimental Guidance and Technical Tips
- The antibody was raised against a recombinant protein corresponding to amino acids 1-116 of human DNAJC19, covering the N-terminal region. It may recognize full-length protein and potential isoforms or cleavage products.
- For Western blotting, a band of approximately 12 kDa is expected. Consider using appropriate molecular weight markers and optimized lysis buffers for mitochondrial protein extraction.
- In immunofluorescence or immunocytochemistry, mitochondrial localization should be observed. Co-staining with mitochondrial markers is recommended for validation.
- Cross-reactivity with mouse and rat DNAJC19 is documented; however, validation in the relevant sample system is advised when using these species.
- ELISA applications may require titration of the antibody and optimization of the detection system.
