Biological Background: DNAJB9 Function and Localization
- DNAJB9, also known as ERdj4 or MDG1, is a member of the DnaJ homolog subfamily B, encoded by the DNAJB9 gene.
- It functions as a co-chaperone for Hsp70 protein HSPA5/BiP, stimulating its ATPase activity and enabling efficient substrate recognition. Related references: PMID:18400946
- As a key repressor of the ERN1/IRE1-mediated unfolded protein response (UPR), DNAJB9 recruits HSPA5/BiP to inactivate ERN1/IRE1 in the unstressed endoplasmic reticulum.
- DNAJB9 is involved in endoplasmic reticulum-associated degradation (ERAD) of misfolded proteins, contributing to protein quality control.
- The protein is localized in the endoplasmic reticulum lumen.
- DNAJB9 is required for survival of B-cell progenitors and normal antibody production.
- Widely expressed, with highest levels in liver, placenta, and kidney. Related references: PMID:11836248
- Keywords: chaperone, endoplasmic reticulum, phosphoprotein, unfolded protein response.
Experimental Guidance and Technical Tips
- The recombinant immunogen corresponds to amino acids 24-223 of human DNAJB9; this region is conserved in mouse and rat, supporting the reported cross-reactivity. Researchers should validate performance in their specific experimental systems.
- For Western blot, a band near 26 kDa is anticipated based on the computed molecular weight. Optimal antibody dilution and incubation conditions should be determined empirically.
- ELISA applications may require optimization of coating antigen and detection conditions; consider testing a range of antibody concentrations.
