Biological Background: DNAJB12 Function and Localization
- DNAJB12 (DnaJ homolog subfamily B member 12), also known as DJ10, is a member of the DnaJ/Hsp40 family of co-chaperones.
- Acts as a co-chaperone with HSPA8/Hsc70, promoting protein folding and trafficking, preventing aggregation, and targeting unfolded proteins to the ERAD pathway. Related references: PMID:21148293, PMID:21150129
- Together with DNAJB14, functions as a chaperone for the maturation of potassium channels KCND2 and KCNH2 by stabilizing nascent subunits and facilitating their oligomerization, independently of HSPA8/Hsc70. Related references: PMID:27916661
- Overexpression of DNAJB12 leads to formation of nuclear membranous structures (DJANGOs) with DNAJB14 and HSPA8/Hsc70, whose cellular role remains unclear. Related references: PMID:24732912
