Biological Background: DNAJA1 Function and Localization

  • DNAJA1, also known as HDJ2 or heat shock 40 kDa protein 4, is a member of the DnaJ/Hsp40 chaperone family. It functions as a co-chaperone for HSPA8/Hsc70, stimulating its ATP hydrolysis activity (PubMed:10816573). Related references: PMID:10816573
  • In its role as a co-chaperone, DNAJA1 is involved in protein transport into mitochondria and assists in the folding and translocation of mitochondrial proteins.
  • DNAJA1 negatively regulates the translocation of the pro-apoptotic protein BAX from the cytosol to mitochondria, protecting cells against apoptosis under stress conditions (PubMed:14752510). Related references: PMID:14752510
  • Conversely, DNAJA1 can also promote apoptosis in response to specific cellular stresses, such as exposure to anisomycin or UV radiation, highlighting its dual role in cell fate decisions (PubMed:24512202). Related references: PMID:24512202
  • DNAJA1 localizes to multiple subcellular compartments, including the cytoplasm, nucleus, mitochondria, endoplasmic reticulum (microsome), and membrane, consistent with its diverse chaperone functions.
  • The protein undergoes various post-translational modifications, including acetylation, methylation, phosphorylation, and prenylation, and contains conserved zinc-finger domains that mediate interactions with client proteins.
  • Isoform 2 of DNAJA1, generated by alternative splicing, shows tissue-specific expression with high levels in testis and lung, while isoform 1 is ubiquitously expressed.

Experimental Guidance and Technical Tips

  • The immunogen was a recombinant fusion protein encompassing amino acids 178–397 of human DNAJA1 (NP_001530.1). This region includes the C-terminal domain and may contribute to recognizing both isoforms; validation in relevant sample systems is recommended.
  • For Western blot (WB), the observed molecular weight is ~45 kDa. Use appropriate controls, including knockout validation if possible, to confirm specificity.
  • In immunofluorescence (IF/ICC) and ELISA applications, optimize antibody dilution and fixation/permeabilization conditions based on the target's subcellular localization (cytoplasmic, nuclear, mitochondrial).
  • As DNAJA1 expression may change under stress conditions, consider using positive control samples treated with heat shock or anisomycin to verify reactivity.
