Biological Background: DNA Polymerase gamma Function and Localization
- DNA Polymerase gamma is the catalytic subunit of the mitochondrial DNA polymerase, responsible for the replication of mitochondrial DNA (mtDNA). It replicates both heavy and light strands using single-stranded DNA templates and RNA primers. Related references: PMID:11477093, PMID:11897778, PMID:15917273
- Possesses 5'->3' DNA polymerase activity and 3'->5' exonucleolytic proofreading activity, ensuring high fidelity during mtDNA synthesis. It proficiently corrects base substitutions, single-base additions, and deletions. Related references: PMID:10827171, PMID:11477094, PMID:11504725
- Forms a highly processive replisome with the mitochondrial helicase TWNK and single-stranded DNA-binding protein SSBP1 at the replication fork. Related references: PMID:11477093, PMID:11897778, PMID:15167897
- Exhibits 5'-deoxyribose phosphate (dRP) lyase activity, mediating repair-associated mtDNA synthesis in the base-excision repair pathway. Related references: PMID:9770471
- Localizes to the mitochondrial matrix and mitochondrial nucleoid, where mtDNA replication and maintenance occur.
- Associated with a spectrum of mitochondrial disorders, including progressive external ophthalmoplegia, Leigh syndrome, and Alpers syndrome, highlighting its critical role in mtDNA stability.
