Biological Background: DNA Polymerase beta Function and Localization
- DNA polymerase beta (POLB), also known as 5'-deoxyribose-phosphate lyase (5'-dRP lyase) or AP lyase, is a repair polymerase that plays a key role in base-excision repair (BER). This pathway corrects damaged bases, and POLB is essential for the repair synthesis step. Related references: PMID:10556592, PMID:9207062, PMID:9572863
- Following base excision and strand nicking by an AP endonuclease, POLB removes the 5'-deoxyribose-phosphate group from the pre-incised AP site via its 5'-dRP lyase activity. It then fills the single-nucleotide gap through its DNA polymerase activity. Related references: PMID:10556592, PMID:17526740, PMID:8841119
- POLB also exhibits AP lyase activity, cleaving sugar-phosphate bonds 3' to an intact abasic (AP) site, thereby contributing to alternative repair pathways. Related references: PMID:9614142
- The polymerase activity of POLB is distributive rather than processive, adding one nucleotide at a time in a stepwise manner, which is distinct from replicative DNA polymerases.
- POLB localizes to both the nucleus and cytoplasm, consistent with its roles in nuclear DNA repair and possible cytoplasmic functions.
- Post-translational modifications of POLB include acetylation, methylation, and ubiquitin conjugation, which may regulate its stability, activity, or interaction with other BER proteins.
- POLB is a key component of the cellular response to DNA damage, contributing to genome stability and playing roles in processes such as DNA replication and repair.
Experimental Guidance and Technical Tips
- The immunogen spans the full-length human POLB protein (amino acids 1–335), which may elicit antibodies against multiple epitopes. Consider this broad reactivity when interpreting results.
- For Western blot, a specific band around 38 kDa is expected. Validate with appropriate positive and negative controls, and consider using KO-validated lysates to confirm specificity.
- This antibody is suitable for IHC-P, IF/ICC, and ELISA. Optimize dilutions and antigen retrieval conditions experimentally for each application and sample type.
- Cross-reactivity with mouse POLB is reported, enabling studies in mouse models. However, verify cross-reactivity in your specific experimental system.
- As a polyclonal antibody, lot-to-lot consistency should be monitored. Include internal reference standards for quantitative comparisons across experiments.
