Biological Background: DNA Polymerase beta Function and Localization
- DNA Polymerase beta (POLB) is a repair polymerase essential for base-excision repair (BER). It possesses 5'-deoxyribose-phosphate lyase (dRP lyase) activity to remove the 5'-dRP group from preincised AP sites, and DNA polymerase activity to fill single-nucleotide gaps with the correct nucleotide. Related references: PMID:10556592 PMID:9207062 PMID:9572863
- POLB conducts gap-filling DNA synthesis in a stepwise distributive manner, adding one nucleotide per binding event, unlike processive polymerases. Related references: PMID:17526740 PMID:8841119
- Beyond its polymerase and dRP lyase activities, POLB can also act as an AP lyase, cleaving sugar-phosphate bonds 3' to an intact abasic site. Related references: PMID:9614142
- Subcellular localization includes both the nucleus and cytoplasm, consistent with its role in nuclear DNA repair and potential cytoplasmic DNA damage responses.
- Post-translational modifications include acetylation, methylation, and ubiquitin-like conjugation, which may regulate its stability and function.
- As a key player in the DNA damage response, POLB is implicated in maintaining genomic stability, and its deficiency is associated with increased sensitivity to alkylating agents.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide mapping to amino acids 236–335 of human POLB; antibody performance should be validated in the specific sample system and application of interest.
- For Western blotting, a band at approximately 38 kDa is expected; consider using appropriate positive and negative controls.
- IHC-P experiments may require optimized antigen retrieval protocols; preliminary testing on positive control tissues (e.g., human tonsil) is recommended.
- The antibody is suitable for ELISA; it is advisable to determine optimal working dilutions by titration.
