Biological Background: DNA-PKcs (DNA-dependent protein kinase catalytic subunit) Function and Localization
- DNA-PKcs is the catalytic subunit of the DNA-dependent protein kinase (DNA-PK) complex, encoded by the PRKDC gene (also known as HYRC, HYRC1, DNPK1, p460).
- As a molecular sensor for DNA damage, DNA-PKcs is essential for non-homologous end joining (NHEJ), the primary pathway repairing double-strand breaks (DSBs) and V(D)J recombination. Related references: PMID:11955432, PMID:12649176, PMID:14734805
- DNA-PKcs requires DNA binding for catalytic activation; it is recruited to DNA ends by the Ku70/Ku80 heterodimer (XRCC6/XRCC5) to protect and align broken DNA ends and sequester the break for NHEJ. Related references: PMID:11955432, PMID:12649176, PMID:14734805
- Beyond DNA repair, DNA-PKcs acts as a scaffold protein, modulating transcription, promoting ribosome assembly by processing precursor rRNA, and maintaining telomeric stability. Related references: PMID:11955432, PMID:12649176, PMID:14734805, PMID:32103174
- DNA-PKcs is primarily localized in the nucleus and nucleolus, with additional presence in the cytosol.
- As a serine/threonine kinase recognizing the [ST]-Q consensus, DNA-PKcs phosphorylates key substrates including H2AX (Ser-139) for DNA damage signaling, AKT (Ser-473) for PI3K/AKT pathway regulation, and numerous DNA repair factors (e.g., XRCC4, XRCC5, XRCC6). Related references: PMID:11955432, PMID:12649176, PMID:14734805, PMID:14627815, PMID:16046194, PMID:15262962
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 3987-4086 of human DNA-PKcs; expect reactivity near the C-terminal region of the protein.
- For Western blot (WB), the predicted molecular weight is approximately 469 kDa; consider using low-percentage SDS-PAGE gradient gels and extended transfer times to ensure efficient blotting of this large protein.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), validation of antigen retrieval methods and fixation conditions in relevant tissue or cell samples is recommended.
- As a polyclonal antibody, lot-to-lot consistency should be confirmed by the supplier; always include appropriate positive and negative controls in your experimental system.
