• DMP1 (dentin matrix acidic phosphoprotein 1), encoded by the DMP1 gene, is also known as DMP-1, ARHP, and ARHR.
• It has a dual role in osteoblast differentiation: in the nucleus of undifferentiated osteoblasts, the unphosphorylated form functions as a transcriptional activator for osteoblast-specific genes such as osteocalcin.
• Upon transition to osteocytes, DMP1 becomes phosphorylated and is exported to the extracellular matrix, where it regulates hydroxyapatite crystal nucleation, contributing to biomineralization.
• Subcellular localization includes the nucleus, cytoplasm, and secretion into the extracellular matrix.
• DMP1 is expressed in tooth tissues, particularly in odontoblasts, ameloblasts, and cementoblasts.
