Biological Background: DLST Function and Localization
- DLST (dihydrolipoamide S-succinyltransferase) is the E2 component of the 2-oxoglutarate dehydrogenase complex, also known as OGDC-E2 or E2K. It catalyzes the transfer of a succinyl group from dihydrolipoamide to coenzyme A, forming succinyl-CoA.
- The 2-oxoglutarate dehydrogenase complex converts 2-oxoglutarate to succinyl-CoA and CO2, a critical step in the tricarboxylic acid (TCA) cycle. Related references: PMID:29211711, PMID:30929736
- A fraction of DLST localizes to the nucleus, where it associates with chromatin and provides succinyl-CoA to histone succinyltransferase KAT2A, facilitating histone succinylation. Related references: PMID:29211711
- The protein is predominantly mitochondrial (matrix) but is also found in the nucleus. It contains lipoyl domains essential for its enzymatic function.
- DLST undergoes post-translational modifications including acetylation and phosphorylation, which may modulate its activity and interactions.
- Alternative splicing produces multiple isoforms, potentially allowing tissue-specific regulation or distinct functional roles.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 150–250 of human DLST. For Western blot, expect a band near 49kDa; validate in human cell lines known to express DLST, such as HeLa or HEK293.
- In IF/ICC and IHC-P, anticipate both mitochondrial and nuclear staining patterns due to the dual localization of DLST. Use appropriate organelle markers for confirmation.
- For ELISA, consider coating with recombinant DLST protein or validated peptide; cross-reactivity is limited to human, so test assay specificity against other species if needed.
- Since this is a monoclonal antibody, lot-to-lot consistency should be high; nevertheless, perform routine quality checks in your experimental system.
