Biological Background: DLK1 Function and Localization
- Protein delta homolog 1 (DLK1), also known as pG2 or PREF1, is a transmembrane protein containing multiple EGF-like repeats, a signal peptide, and a single transmembrane helix.
- It is expressed in stromal cells of placental villi, yolk sac, fetal liver, adrenal cortex, pancreas, and in beta cells of the islets of Langerhans; also detected in some neuroendocrine lung tumor tissues.
- DLK1 may play a role in neuroendocrine differentiation.
- The protein undergoes alternative splicing, generating multiple isoforms, and is post-translationally modified by glycosylation and disulfide bond formation.
- Subcellular localization includes the cell membrane and cytoplasm.
- The protein is also known by aliases such as Delta1 and FA1, reflecting its identification in various developmental and tumor contexts.
- 3D structural data and direct protein sequencing are available, supporting its classification as a reference proteome member.
Experimental Guidance and Technical Tips
- The immunogen (aa 24–303) covers most of the extracellular region, including EGF-like domains, which may facilitate detection of both membrane-bound and shed forms of DLK1.
- For Western blot, consider using both reducing and non-reducing conditions to account for the disulfide-stabilized EGF-like repeats; the predicted molecular weight is ~41 kDa, but glycosylation may shift migration.
- In IHC-P, validate in known positive tissues such as placenta or pancreas, and include neuroendocrine tumor samples if relevant; antigen retrieval conditions should be optimized.
- For IF/ICC, permeabilization is recommended to detect cytoplasmic localization; confirm specificity using knockout or knockdown cell lines where possible.
- Cross-reactivity with mouse and rat is reported; verify performance in the specific species and sample system of interest.
- As a polyclonal antibody, it may recognize multiple isoforms and degradation products; use appropriate controls (e.g., peptide blocking, isotype controls) to confirm target bands.
