Biological Background: DLDH/DLD Function and Localization
- DLD functions as a critical E3 component in the pyruvate dehydrogenase, alpha-ketoglutarate dehydrogenase, and branched-chain amino acid dehydrogenase complexes, catalyzing reoxidation of the dihydrolipoyl moiety on E2 with NAD+ as the electron acceptor. Experimental Guidance and Technical Tips
- The immunogen was designed using a recombinant fragment spanning amino acids 36–300 of human DLD, which may predominantly target the N-terminal domain. Consider this when interpreting band patterns or cross-reactivity.
- For Western blot, the expected molecular weight is ~54 kDa. Validate band specificity in the relevant sample system, as post-translational modifications or isoforms may produce additional bands.
- In IHC-P, optimize antigen retrieval conditions and antibody dilution. Endogenous peroxidase and biotin may need to be blocked depending on the detection system.
- ELISA performance can be influenced by coating antigen purity and format. Titrate the antibody in the appropriate assay buffer to establish working concentrations.
- This antibody shows cross-reactivity with mouse, making it suitable for studies in mouse models. Species cross-reactivity beyond human and mouse has not been specified; independent validation is recommended.
