Biological Background: DLC1 Function and Localization
- DLC1 is a Rho GTPase-activating protein (RhoGAP) that specifically inactivates the small GTPases RHOA, RHOB, RHOC, and CDC42, thereby terminating their downstream signaling cascades.
- It plays a critical role in cytoskeletal reorganization, cell migration, and proliferation, and is recognized as a tumor suppressor in multiple cancer types.
- In addition to its RhoGAP activity, DLC1 functions as an activator of phospholipase PLCD1.
- During growth factor-induced epithelial cell migration, DLC1 forms complexes with TNS3 and PTEN; upon stimulation, phosphorylation triggers a switch where PTEN binds DLC1 and TNS3 binds p85, leading to RHOA activation at the cell posterior and RAC1 activation at the leading edge. - In ELISA applications, optimize antigen coating conditions using recombinant full-length DLC1 or the immunogen fragment and verify linearity of detection with serial dilutions of standard protein.
