  • DLAT encodes the dihydrolipoyllysine-residue acetyltransferase component of the pyruvate dehydrogenase complex (PDC-E2). It is also known as the 70 kDa mitochondrial autoantigen of primary biliary cirrhosis (PBC), M2 antigen complex 70 kDa subunit, and PDCE2.
  • As part of the pyruvate dehydrogenase (PDH) complex, DLAT catalyzes the transfer of acetyl groups from acetyl-lipoyl moieties to coenzyme A, forming acetyl-CoA. This reaction links cytoplasmic glycolysis to the mitochondrial tricarboxylic acid (TCA) cycle.
  • The PDH complex is a large multienzyme assembly containing multiple copies of three enzymes: pyruvate dehydrogenase (E1), dihydrolipoamide acetyltransferase (E2), and dihydrolipoamide dehydrogenase (E3). DLAT functions as the E2 component.
  • DLAT localizes to the mitochondrial matrix, where the PDH complex is active.
  • The protein undergoes lipoylation and acetylation, post-translational modifications essential for its enzymatic function and regulation.
  • DLAT is a transferase that plays a key role in carbohydrate and glucose metabolism, specifically in the TCA cycle.

  • Validate reactivity in the relevant species: human, mouse, and rat cell lines or tissue extracts. Include appropriate positive and negative controls.
