Biological Background: Dickkopf-related protein 3 Function and Localization

  • Dickkopf-related protein 3 (DKK3), encoded by the DKK3 gene (also known as REIC), is a member of the dickkopf family of secreted Wnt antagonists.
  • DKK3 antagonizes canonical Wnt signaling by inhibiting the interaction between LRP5/6 co-receptors and Wnt ligands, and also promotes LRP5/6 internalization through complex formation with KREMEN.
  • During vertebrate development, DKK3 locally inhibits Wnt-regulated processes including antero-posterior axial patterning, limb development, somitogenesis, and eye formation.
  • In adults, DKK3 is implicated in bone formation, bone disease, cancer, and Alzheimer's disease.
  • DKK3 is a secreted protein, with a signal peptide, and is post-translationally modified by glycosylation and disulfide bond formation; it also contains a coiled-coil domain.
  • The protein shows highest expression in heart, brain, and spinal cord.

  • In immunofluorescence/immunocytochemistry, as DKK3 is secreted, consider using secretion inhibitors or staining under conditions that retain extracellular matrix-bound protein; validate in relevant cell types.
  • When using this antibody for ELISA, the immunogen covers the N-terminal region (aa 22-200), which may recognize DKK3 even in complex with other proteins; perform sandwich ELISA optimization.
