Biological Background: DKK3 Function and Localization
- Dickkopf-related protein 3 (DKK3), also known as REIC, RIG, or CRRL, is a secreted glycoprotein belonging to the dickkopf family.
- DKK3 functions as an antagonist of the canonical Wnt signaling pathway by inhibiting the interaction of LRP5/6 with Wnt ligands and by forming a ternary complex with KREMEN to promote internalization of LRP5/6.
- DKK3 plays a critical role in vertebrate development, locally inhibiting Wnt-regulated processes such as antero-posterior axial patterning, limb development, somitogenesis, and eye formation (By similarity).
- In adult tissues, DKK3 is implicated in bone formation, bone disease, cancer, and Alzheimer disease (By similarity).
- DKK3 contains a signal peptide, a coiled-coil domain, and multiple disulfide bonds, and is subject to glycosylation.
- Highest expression of DKK3 is observed in heart, brain, and spinal cord.
- Subcellularly, DKK3 is secreted into the extracellular space.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 22–200 of human DKK3 (NP_056965.3), spanning the N-terminal extracellular domain after signal peptide cleavage. This region includes conserved cysteine-rich domains critical for Wnt interaction; thus, the antibody may recognize both native and denatured protein conformations.
- Suitable applications validated by the supplier include WB, IF-P, and ELISA. For WB, a band of approximately 38 kDa is expected. Consider using a reducing agent to break disulfide bonds if necessary.
- Detects human, mouse, and rat DKK3; cross-reactivity with other species should be verified experimentally.
- For immunohistochemistry/immunofluorescence (IF-P), antigen retrieval may be required to expose epitopes in formalin-fixed tissues. Validate in the relevant sample system.
