Biological Background: DIMT1 Function and Localization

  • DIMT1 (Dimethyladenosine transferase) is an S-adenosylmethionine-dependent methyltransferase that catalyzes N6-dimethylation of two adjacent adenosines (A1779 and A1780) in a conserved hairpin loop near the 3′ end of 18S rRNA within the small ribosomal subunit.
    Related references: PMID:25851604
  • Beyond its catalytic activity, DIMT1 participates in pre-rRNA processing and maturation of the small ribosomal subunit; this function is independent of its methyltransferase activity.
    Related references: PMID:25851604
  • DIMT1 is an integral component of the small subunit (SSU) processome, the earliest precursor of the small eukaryotic ribosomal subunit, where it cooperates with numerous ribosome biogenesis factors, RNA chaperones, and ribosomal proteins to ensure proper pre-rRNA folding, modification, cleavage, and quality control via the RNA exosome.
    Related references: PMID:34516797
  • The protein is also known as DIM1 dimethyladenosine transferase 1 homolog, DIMT1L, HUSSY5, and HSA9761; it is the human ortholog of yeast Dim1p.
  • DIMT1 localizes to the nucleus, with predominant accumulation in the nucleolus, consistent with its role in early ribosome assembly.
  • It possesses RNA-binding capacity and belongs to the methyltransferase superfamily, utilizing S-adenosyl-L-methionine (SAM) as a methyl donor.
  • Structural studies (3D-structure available) have provided insights into its catalytic mechanism and interactions with RNA substrates.

Experimental Guidance and Technical Tips

  • The immunogen used for this polyclonal antibody corresponds to the N-terminal 1–126 region of human DIMT1. This region is likely to be conserved across species, as indicated by the reported cross-reactivity with mouse and rat.
  • For Western blot analysis, a band at approximately 35 kDa is expected in lysates from human, mouse, or rat cell lines; however, optimal sample preparation and detection conditions should be empirically determined.
  • In ELISA applications, the antibody can be used as a capture or detection reagent; titration and appropriate blocking steps are recommended to achieve optimal signal-to-noise ratios.
  • As this antibody may recognize both the methyltransferase-active and processing-related conformations of DIMT1, it is suitable for studying ribosome biogenesis and rRNA modification pathways.
