Biological Background: DNA (cytosine-5)-methyltransferase 3A (DNMT3A) Function and Localization

  • DNMT3A is a key enzyme required for genome-wide de novo DNA methylation, essential for establishing methylation patterns during development. It modifies DNA in a non-processive manner and can methylate both CpG and non-CpG sites.
  • The enzyme functions in coordination with histone modifications, preferentially methylating linker DNA between nucleosomes and is inhibited by histone H1. It also plays a role in paternal and maternal imprinting, with methylation of most imprinted loci in germ cells.
  • DNMT3A acts as a transcriptional corepressor, for example, by interacting with ZBTB18, and can repress transcription via recruitment of HDAC activity. It is recruited to genomic regions marked by trimethylated histone H3 at Lys36 (H3K36me3).
  • Subcellularly, DNMT3A localizes predominantly in the nucleus and associates with chromosomes, accumulating at pericentric heterochromatin major satellite repeats. It may also be found in the cytoplasm.
  • Two isoforms exist: isoform 1 is ubiquitously expressed at low levels, while isoform 2 expression is restricted to tissues undergoing active de novo methylation, such as spleen, testis, and thymus.
  • The protein undergoes multiple post-translational modifications, including phosphorylation (e.g., at Ser102, Ser239, Ser251, Ser386, Ser389, Thr120, Thr257), methylation (e.g., omega-N-methylarginine at Arg167), and auto-methylation on Cys706.
