- DICER1 is a ~219 kDa endoribonuclease belonging to the RNase III family, also known as Helicase with RNase motif.
- It plays a central role in RNA interference by cleaving long double-stranded RNA (dsRNA) into small interfering RNAs (siRNAs) and processing precursor microRNAs into mature miRNAs, each approximately 21-23 nucleotides long.
- These small RNAs are loaded into the RNA-induced silencing complex (RISC), guiding sequence-specific degradation or translational repression of target mRNAs.
- DICER1 localizes primarily to the cytoplasm, with enrichment in the perinuclear region.
- The protein contains conserved helicase and RNase III domains, as well as an ATP-binding site, and requires divalent metal ions such as magnesium or manganese for catalytic activity.
- Post-translational modifications, including phosphorylation, and alternative splicing generate multiple DICER1 isoforms, which may have distinct regulatory roles.
