Biological Background: DIAPH1 Function and Localization
- DIAPH1, also known as protein diaphanous homolog 1 or diaphanous-related formin-1 (DRF1), is encoded by the DIAPH1 gene in humans. It belongs to the formin family of actin nucleation factors.
- Functions as an actin nucleation and elongation factor essential for the assembly of F-actin structures, including actin cables and stress fibers. It binds to barbed ends of actin filaments and modulates polymerization dynamics. Related references: PMID:20937854 PMID:21834987
- Acts as a scaffold protein linking the actin and microtubule cytoskeletons by interacting with MAPRE1 and APC to stabilize microtubules and promote cell migration. Related references: PMID:20937854 PMID:21834987
- Plays a critical role in Rho- and Src-dependent signaling pathways, coupling extracellular cues to cytoskeletal remodeling, and is required for cytokinesis, serum response factor (SRF) transcriptional activation, and cell shape control.
- Localizes to multiple subcellular compartments including the cell membrane, ruffles, centrosome, spindle, cytoplasm, and nucleus, reflecting its diverse roles in organizing the cytoskeleton.
- Highly expressed in brain, heart, placenta, lung, kidney, pancreas, liver, skeletal muscle, and cochlea, with notable presence in platelets, consistent with its involvement in cell motility and hearing. Related references: PMID:26912466
- In hair cells, DIAPH1 regulates actin polymerization, which is crucial for auditory function; defects lead to DFNA1 non-syndromic sensorineural hearing loss. Related references: PMID:20937854 PMID:21834987 PMID:26912466
- Additionally, DIAPH1 promotes nuclear actin polymerization, driving serum-dependent SRF-MRTFA transcriptional activity (By similarity).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 1-100 of human DIAPH1; this N-terminal region may target a conserved epitope, likely contributing to the observed cross-reactivity with mouse and rat.
- For Western blotting, the predicted molecular weight is approximately 141 kDa. Consider using a low-percentage gel and adequate transfer conditions for high-molecular-weight proteins.
- In ELISA applications, the antibody may be used as a capture or detection reagent; however, optimal working concentrations should be empirically determined.
- Given its widespread expression, validate detection specificity in relevant cell lines (e.g., HeLa, HEK293, NIH/3T3) or tissue lysates, ensuring appropriate positive and negative controls.
- For cytoskeletal studies, consider combining this antibody with phalloidin staining to correlate DIAPH1 localization with F-actin structures.
