Biological Background: DIAPH1 Function and Localization
- DIAPH1 (Protein diaphanous homolog 1) is a diaphanous-related formin that functions as an actin nucleation and elongation factor, essential for cytoskeletal organization.
- Required for the assembly of F-actin structures (stress fibers, cables), cytokinesis, and serum response factor (SRF) transcriptional activation (By similarity).
- Acts as a scaffold for MAPRE1 and APC to stabilize microtubules, promoting cell migration and neurite outgrowth (By similarity).
- In hair cells, regulates actin polymerization, a process crucial for hearing. Related references: PMID:20937854 / PMID:21834987 / PMID:26912466
- The MEMO1-RHOA-DIAPH1 pathway is involved in ERBB2-dependent microtubule stabilization at the cell cortex and modulates GSK3B to control APC/CLASP2 localization. Related references: PMID:20937854 / PMID:21834987
- Plays a role in brain development. Related references: PMID:24781755
- Subcellular localization includes the cell membrane, ruffles, cytoplasm, cytoskeleton, centrosome, spindle, and nucleus.
- Expressed in many tissues, including brain, heart, placenta, lung, kidney, pancreas, liver, skeletal muscle, cochlea, and platelets. Related references: PMID:26912466
- Post-translational modifications include acetylation and phosphorylation (UniProt keywords: Acetylation, Phosphoprotein).
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1158–1272 (C-terminal region) of human DIAPH1, which may recognize full-length protein and specific isoforms.
- For Western blot, expect a band at approximately 141 kDa in human cell lysates; validate the detection in relevant samples such as HeLa or HEK293.
- This polyclonal antibody is suitable for ELISA; consider optimizing coating antigen and detection conditions for indirect or sandwich formats.
- Cross-reactivity is validated in human; for non-human species, perform a pilot experiment to confirm recognition.
- The [KD Validated] designation indicates knock-down validation in published studies; always confirm specificity in your own experimental system.
