- DHX58, also known as LGP2 or RLR-3, is an ATP-dependent RNA helicase and a member of the RIG-I-like receptor (RLR) family. It lacks the CARD domain required for direct MAVS activation, serving as a regulator rather than an initiator of antiviral signaling.
- DHX58 exerts dual regulatory roles on RIG-I and MDA5 signaling: it can negatively regulate by competing for viral RNA or binding to RIGI and preventing its interaction with MAVS, and positively regulate by unwinding viral nucleoproteins to facilitate their recognition by RIG-I and MDA5.
- It is involved in the innate immune response to a broad range of pathogens, including RNA viruses, DNA viruses (such as poxviruses and SARS-CoV-2), and the bacterium Listeria monocytogenes. Related references: PMID:31256877
- DHX58 binds both single-stranded RNA (ssRNA) and double-stranded RNA (dsRNA) with higher affinity for dsRNA, and shows a preference for 5'-triphosphorylated RNA.
