Biological Background: DHX30 Function and Localization
- DHX30 (DEAH-box helicase 30), also known as DDX30, is an ATP-dependent RNA helicase belonging to the DEAH-box protein family.
- It plays a critical role in mitochondrial ribosome large subunit assembly, essential for mitochondrial protein synthesis (PubMed:25683715, PubMed:29100085). Related references: PMID:29100085 PMID:25683715
- DHX30 associates with mitochondrial DNA and localizes to the mitochondrial nucleoid, suggesting involvement in mitochondrial DNA organization (PubMed:18063578). Related references: PMID:18063578
- It is required for optimal function of the zinc-finger antiviral protein ZC3HAV1, implicating a role in innate immune responses (By similarity).
- DHX30 contributes to nervous system development and differentiation by up-regulating neurogenesis-related genes (By similarity).
- The protein predominantly localizes to the mitochondrial matrix and nucleoid, as well as the cytoplasm.
- Post-translational modifications include phosphorylation, as identified by proteomics, and the gene undergoes alternative splicing.
- Disease-associated mutations in DHX30 are linked to neurodevelopmental disorder with mitochondrial dysfunction (NEDMIAL) and intellectual disability.
Experimental Guidance and Technical Tips
- The antibody was raised against a recombinant protein covering amino acids 336–501 of human DHX30, a region likely accessible for antibody binding. Confirm specificity in your assay system.
- For Western blotting, the expected molecular weight is approximately 134 kDa; use appropriate molecular weight markers and validate lysate quality.
- In ELISA applications, the recombinant immunogen can serve as a positive control to optimize assay conditions.
- Due to reported cross-reactivity with mouse, this antibody may be suitable for studies in mouse models; however, validate in the relevant sample system.
