Biological Background: DHRS2 Function and Localization
- DHRS2, also known as HEP27 or SDR25C1, is a member of the short chain dehydrogenase/reductase (SDR) family. It is encoded by the DHRS2 gene, with alternative splicing producing multiple transcript variants.
- NADPH-dependent oxidoreductase that reduces dicarbonyl compounds, playing a role in inactivation of reactive carbonyls. Shows reductase activity with substrates like 3,4-hexanedione, 2,3-heptanedione, and 1-phenyl-1,2-propanedione. Related references: PMID:16685466
- Exhibits minor hydroxysteroid dehydrogenase activity toward bile acids, but unlikely to control hormone levels. Does not act on retinoids or sugars. Related references: PMID:16685466
- Attenuates MDM2-mediated degradation of p53/TP53, leading to p53 stabilization and increased transcription activity, resulting in accumulation of MDM2 and CDKN1A/p21. This suggests a tumor-suppressive role. Related references: PMID:20547751
- Reduces proliferation, migration, invasion, and ROS production in cancer cells. Related references: PMID:29106393
- Subcellular localization includes the mitochondrial matrix and the nucleus, as per UniProt annotation.
- Widely expressed, with highest levels in liver and kidney; also detected in heart, spleen, skeletal muscle, placenta, and dendritic cells, but not in many other immune cells.
- Post-translational modifications include acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- For Western blotting, the antibody targets a ~30 kDa band. Validate specificity in DHRS2-expressing tissues such as liver or kidney, and consider using positive controls (e.g., HeLa or HepG2 cell lysates).
- In immunofluorescence/ICC, the antibody may label mitochondria and nuclei. Optimize fixation and permeabilization conditions to preserve epitope accessibility.
- For ELISA applications, the polyclonal nature may enhance detection sensitivity. Pair with a monoclonal detection antibody if developing a sandwich assay.
- Cross-reactivity with mouse and rat homologs is indicated; verify sequence homology before applying to these species.
