Biological Background: Diacylglycerol kinase zeta Function and Localization
- Diacylglycerol kinase zeta (DGK-zeta), encoded by the DGKZ (DAGK6) gene, is a kinase with ANK repeats and zinc-finger domains.
- Converts diacylglycerol (DAG) to phosphatidic acid (PA), regulating the balance of these bioactive lipids and acting as a switch between their signaling pathways. Related references: PMID:15544348, PMID:18004883, PMID:19744926
- Plays a role in the biosynthesis of complex lipids (Probable).
- Exhibits no acyl chain-dependent substrate specificity among diacylglycerol species but can phosphorylate 1-alkyl-2-acylglycerol in vitro with a preference for arachidonoyl-containing species. Related references: PMID:9159104, PMID:15544348, PMID:19744926, PMID:22108654, PMID:22627129
- Negatively regulates T-cell receptor signaling, modulating T cell activation (By similarity).
- Stimulates PIP5KIA activity to promote actin polymerization. Related references: PMID:15157668
- Subcellular localization includes the nucleus, cytoplasm, cytosol, cell membrane, and lamellipodium.
- Tissue specificity: highest expression in brain, with substantial levels in skeletal muscle, heart, and pancreas; predominantly expressed in muscle.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (amino acids 808–1117) of human DGKZ, which may be suitable for detecting full-length and alternatively spliced isoforms.
- For Western blot (WB), expect a band at approximately 104 kDa; validate in relevant human or mouse sample systems.
- For ELISA, consider testing under both direct and indirect formats, optimizing antibody concentration and blocking conditions.
- Note the reported cross-reactivity with mouse; sequence homology should be examined for other species if needed.
- As a polyclonal antibody, lot-to-lot consistency should be verified for quantitative assays.
